产品介绍

      Axygen 迷你型加热封口机,适合封口各种微量板以防止蒸发,污染和冻烧。

      该机采用热焊各种不同种的膜到微量板,细胞培养板,PCR板甚至深孔板。

      数字式温度测控,时间控制,允许用户选择不同的封口膜材料和板的封口参数。

      因有内置的压力传感装置,每个循环,系统可以施加相同的压力150Kg,传递均匀的热量到膜,无论是PCR板还是深孔板,封口稳定。

步骤:

  1. 设定温度
  2. 设定热封时间长短
  3. 预热10分钟,等待温度达到设定值并稳定在设定值
  4. 将待封板放置在正确的封口托盘上,并将膜放置在板上
  5. 用把手将封口托盘推入左边到封口热头下边的正确位置,安全门会自动关闭
  6. 按下绿色开始按钮,热封头自动向下,接触板子,压封。可以看到时间计数在减少,当时间到达零时,热头抬起,离开板子,回到上面原位
  7. 将封好的板子向右推出,取出封好的板子,完成了。(注意可能还很烫哦,允许放置让其冷却下来,在进行其他操作。)

Teklad guinea pig diets are complete and balanced diets formulated to meet the nutritional needs of guinea pigs during various stages of the life cycle.

Protein supplied by plant and fish meal sources provides the balance of essential amino acids required by guinea pigs. Fat from soybean oil supplies essential fatty acids necessary for a good hair coat.

Teklad guinea pig diets are properly fortified with vitamins and minerals to meet the needs of guinea pigs during growth, maintenance, and reproduction.

Vitamin C is included in these diets because guinea pigs (as do man and other primates) require a dietary source of vitamin C to prevent scurvy. Stabilized vitamin C (L-ascorbyl-2-polyphosphate) is used to extend the shelf life.

Alfalfa meal is included as a high-quality forage product to help maintain the normal functioning of the guinea pig’s digestive system. The firm, pelleted diet aids in preventing feed wastage.

显色法检测组胺试剂盒

  组胺多数产生于金枪鱼,鲣鱼,鲭鱼此类海鲜的腐败过程中。

  对于以生鱼(鲜鱼、冷冻鱼肉)为原料的食品来说,为了防止组胺过量而导致过敏性中毒,对于食品组胺浓度测定的管理是很重要的。

  但是,以前的组胺检测需要耗时一天,检测费用高,需要使用大型检测设备。如果用显色法组胺检测系统,生鱼(鲜鱼、冷冻鱼肉)的组胺含量检测可以快捷地完成。

◆特点

显色法检测组胺试剂盒

  ● 简单: 前处理操作简单,可以省略荧光光谱法和高效液相色谱法那样测定障碍物质复杂的前处理操作。由于酶的特异性高,无需使用强酸或有机溶剂,任何对象均可简单、安全地分析

  ● 迅速 利用具有高组胺特异性的组胺脱氢酶的酶法来检测。如果是生鱼,检测和提取过程仅需要1个小时即可得到结果。

◆测定原理


  组胺含有1-Methoxy PMS的情况下,添加组胺脱氢酶会被特异性分解。此时因产生的电子转移会通过四唑盐显色,便可以在470nm上下测量吸光度,计算组胺浓度。

关于组胺

Q1: 什么是组胺?

A1: 一种由组氨酸(氨基酸)合成的胺。如果红身鱼(金枪鱼,秋刀鱼,鲣鱼,竹荚鱼,青花鱼等)等含有大量

    组氨酸的食物,被组胺菌污染的话,会产生组胺。

    另外,一般来说,人的喉咙和鼻粘膜上皮都存在着很多肥大细胞、嗜碱性粒细胞等。一旦数量过多,也会引

    起过敏症状。

 

Q2: 什么是过敏性食物中毒(组胺中毒)?

A2: 红身鱼被组胺菌污染的原因是,食品在制造加工、运输时,在家庭等地方的保存温度管理不当时,组胺菌繁

    殖增多,产生大量组胺。(参照:Q1.什么是组胺?)

    组胺非常耐热,一旦生成,普通的加热处理无法使它分解,还是会残留在鱼肉里。因此,人们食用了被组胺

    污染的鱼或其加工食品,可能会出现过敏症状。在食品安全学上,这种症状被称为过敏性食物中毒(组胺中

    毒)。


Q3: 食物中含有多少组胺时,会引起食物中毒?

A3: 一般来说,食物的组胺含量超过100mg/100g=1000ppm以上的话,会引起食物中毒。

    组胺是会引起过敏反应,胃酸分泌,血压等各种生理反应的化学物质。因此,摄取高浓度组胺的话,约30分

    钟~60分钟后,会出现面部、特别是嘴巴周围以及耳垂的泛红症状,还会出现头痛,荨麻疹,发热等。一般

    经过6~10小时后会恢复正常。


Q4: 对于食物中含有的组胺量标准,是否有相关规定?

A4: 日本:没有相关法制规定。

    欧美:有相关规定。因此,出口到欧美国家的食品,必须要严格遵守规定。


Q5: 有什么方法可以检测组胺量?

A5: 检测鱼肉中的组胺量方法:生物检测技术,比色法,荧光法,高速色谱法(HPLC),抗原 – 抗体反应和毛

    细管电泳等。

    一般来说,日本是使用卫生检测法的HPLC法;美国则是使用荧光光度法(被记载于国际标准—-AOAC法

    内)。



关于龟甲万的组胺检测法

Q1: 龟甲万的组胺检测法与其他检测法相比有什么优点?

A1: ・提取组胺的方法简单,无需进行除去阻碍检测物质的前处理。

    ・与HPLC法、其他公司的同类产品相比,操作更简单。

    ・可以快速、精准度高地检测组胺。

    ・与HPLC法有着很高的相关性。


Q2: 需要什么仪器或器具?

A2: 必需:组胺检测试剂盒(显色法检测组胺),分光光度计。

    其他:请阅读说明书的第一页右下角(试剂盒所需的器具)。

    注意:请使用塑料器具。因为组胺会附着在玻璃上。


Q3: 一般的分光光度计也可以检测吗?

A3: 可以。但需要是可以检测460nm上下的吸光度的分光光度计。

    注意:推荐使用的分光光度计(分光光度计BABS-B470,共立理化学研究所公司所制)的光路是2cm。

    如果使用光路是1cm的分光光度计,一旦超出组胺的检测界限,标准溶液的检测值也会变化。

关于显色法组胺检测试剂盒

Q1: 显色法检测组胺试剂盒的检测浓度范围?

A1: 使用推荐的分光光度计B:可以检测的组胺浓度是0.4-6ppm。如果是检测生鱼,罐头鱼等,将样品稀

    释25倍,再换算样品的浓度,可以检测组胺的范围是10-150ppm。

    光路长1cm的分光光度计:可以检测的组胺浓度是0.8-12ppm。

    如果是检测生鱼,罐头鱼等,将样品稀释25倍,再换算样品的浓度,可以检测组胺的范围是

    20-300ppm。

    此外,若样本的组胺浓度超过检测范围的话,可以稀释样本后,再检测。


Q2: 1盒显色法组胺检测试剂盒可以检测多少个样本?

A2: 本试剂盒可以检测60次。但是,在实际的检测中,每进行一次连续的检测,就需要检测一次标准值。因

    此根据每次连续检测的样本数的不同,一个试剂盒可以检测的样本数也不同。

    例如:

    如果一次的检测就把试剂盒用完的话,除去检测一次标准值之外,实际可以的样本数为59个。

    如果每次检测的样本数为5个的话,再加上每次都需要检测一次标准值。实际可以检测的次数为10次,即

    可以检测的样本数为50个。

    注意:在一次连续的检测中,将没用完的冷冻、冷藏后的试剂替换成新的试剂时候,请再次重新检测标准

    值。


Q3: 如果没有可以调制组胺提取溶液的设备,可以买其他的代替吗?

A3: 不好意思,无法提供组胺提取液的购买。若调制溶液时遇到问题,可以使用其他替代品,详情请咨询金畔。


Q4: 如何保存溶解后剩下的显色试剂?

A4: 最好可以一次性用完溶解后的显色试剂。

    如果真的用剩了,请在按以下方法进行保存:

    冷藏:2~8℃,可保存一周

    冷冻:-10℃以下,可保存一个月。

    在以上保存的情况下,冻结溶解操作只可进行3次。

    解冻时,请在流水中快速进行。解冻完请一定要冷却后再使用。


Q5: 如何保存溶解后剩下的酶试剂?

A5: 最好可以一次性用完溶解后的酶试剂。

    如果真的用剩了,请在按以下方法进行保存:

    冷冻:-10℃以下,可保存一个月。冻结溶解操作只可进行3次。(详情请阅读说明书第二页左方 “2.试剂

    的调制(酶试剂)”)

    还有,当用完冷冻溶解后的酶试剂后,换成新的酶试剂时,请重新检测标准值。



关于从样本中提取组胺

Q1: 为什么从活鱼提取组胺时,需要进行煮沸处理?

A1: 因为组胺是存在于鱼肉成分(肌肉蛋白、油等)中,所以加热后再提取,效率更高。而罐头类食品本身已经过

    加热处理了,提取时不需要再煮沸。


Q2: 如果不能立刻检测样本,或者是已提取了组胺的样本的话,应该怎样做?

A2: 活鱼:请冷冻保存。保存方法没有什么需要注意的点,不过为避免解冻时的酶反应发生得太快,请尽快完

    成解冻。

    ・经过煮沸提取组胺,再过滤后的样本,可以进行放入冷藏、冷冻格里保存。

    ・组胺提取没有经过煮沸的样本,以及对样本进行前处理的时候,由于样本中含有酶,需要进行冷冻。解

    冻的时候,如果有沉淀物出现的话请用滤纸等进行除去。


Q3: 听说样本中含有杂质时,是不能进行检测的。那么杂质是什么物质?

A3: 影响组胺检测试剂盒反应的杂质是氧化还原物质。

    鱼粉:饲料不新鲜时,由此而增殖的微生物形成的氧化还原物质就是杂质。

    罐头:调味料可能具有氧化还原能力。

 

Q4: 如果样本有杂质,检测值是偏高还是偏低?

A4: 如果还原物质多的话,检测值会偏高,如果氧化物质多的话,检测值会偏低。因此检测值是偏高还是偏低,

    是根据两种物质的比例来决定的。氧化还原物质是抗氧化物质(维生素和细胞色素等)或者是氧化物质,在

    食品中以多种类的方式存在,因此无法确认是哪个种类物质造成的影响。


Q5: 如何知道样本中是否含有杂质?

A5: 对新的食品进行组胺检测时,可以在样本添加定量的组胺,然后进行检测(添加回收实验)。由此可以知道

    是否有杂质的影响,以及是否需要进行前处理。


Q6: 是否有排除杂质影响的方法?

A6: 鱼肉、金枪鱼罐头、鱼酱油等食品,只要按照说明书操作(一般的操作的提取、检测),是可以除去杂质的

    影响。

    但是,如果使用一般方法,也不能排除杂质影响的话,可以通过稀释样品或固相萃取柱将氧化还原物质进行

    处理。(详情请咨询金畔)



◆关于检测值有误的问题

Q1: 标准溶液的数值偏低。

A1: 如果使用了溶解后再保存的酶溶液,因为在保存情况下酶活性降低,标准值可能也会降低。但只要达到正常

    值(0.85)的80%以上,检测就没有问题。

    注意:使用光路为1cm的光度计的时候,检测值达到一半以上即可(0.5左右)。(详情请阅读说明书:第

    页右方检测5)


Q2: 样本的组胺量比实际要低。

A2: ・样本的均化没有很好地进行。

    (如果样本的均化没有很好地进行,将无法完全地提取组胺)

    ・样本中含有杂质(参考Q.如果样本有杂质,检测值是偏高还是偏低?)

 

Q3: 样本的组胺浓度超出检测界限。

A3: 稀释组胺提取液后,再次检测。计算出组胺浓度时,注意要考虑稀释倍数,再进行计算。


Q4: 计算样本的组胺量后,得到负数。

A4: 该样本的组胺量,在检测范围的10ppm以下。(使用推荐的仪器分光光度计B的情况下)

    得到负数的原因:

    样本中不存在组胺。

    组胺浓度在检测范围以下时,因有检测误差,计算样本检测值和样本空白值的差值,得到负数。

    推荐仪器(分光光度计B)的检测范围的下限是10ppm。因此,得到负数是因为,样本的组胺浓度会出现

    10ppm以下。


Q5: 检测完的反应液在放置一阵之后,再次检测后发现,检测值变高。那么,哪个数值才是正确?

A5: 反应结束后的检测值是正确的。

    试剂盒的显色试剂,是通过阳光来显色的。如果把反应液放置在有阳光照射的地方,检测值会变高。因此,

    反应结束后,请迅速进行检测。


Q6: 活的红身鱼在常温下放置了几天。检测后发现,鱼中并不含有组胺。

A6: 红身鱼(金枪鱼,秋刀鱼,鲣鱼,竹荚鱼,青花鱼等)等含有大量的组氨酸的食物,若被组胺菌污染的话,

    会产生组胺。因此,还未被组胺菌污染的红身鱼即使腐烂了,也不会被检出组胺。

ISOGEN

  ISOGEN是从人、动物、植物及细菌提取RNA试剂。可通过液相分离方法从同一样本中分离DNA及蛋白质。可通过一系列的操作分离RNA、DNA、蛋白质,有助于分析珍贵的样本。操作简单,便于样本数量较多的检测。

  适用样本:组织、细胞、植物、细菌

ISOGEN-LS

  ISOGEN-LS是从血液或血清等容量较大(>100 µl:使用1.5 ml塑料(离心)管进行处理),难以浓缩的液体样本中提取RNA的试剂。是将ISOGEN针对液体试剂进行改良。除了加入到样本的ISOGEN-LS量比不一样以外,得到的结果与使用 ISOGEN时一样。

  适用样本: 全血、液体样本

◆原理

  本产品是含有苯酚及硫氰酸胍的均一液体。往样本加入ISOGEN或ISOGEN-LS后使其溶解或均匀化,再加入氯仿进行离心分离,匀浆就会分离成水相、中间相及有机相。水相中只有RNA,DNA及蛋白质存在于中间相及其以下。首先,取出水相,加入异丙醇使RNA沉淀。往剩余的中间相及有机相里加入乙醇使DNA沉淀,加入异丙醇使蛋白质沉淀,从而依次分离出RNA、DNA及蛋白质。


      ISOGEN、ISOGEN-LS

  ・主要成分为苯酚及硫氰酸胍的均一溶液

  ・便于液层分离的染色溶液(ISOGEN : 蓝色、ISOGEN-LS : 红色)

◆原理

       ISOIL For Beads Beating是能够萃取土壤中DNA的试剂盒,使用界面活性剂能够进行化学性的溶菌和Beads Beating的物理性菌体破碎从而萃取出土壤中的DNA。即使有坚固细胞壁的微生物中也能萃取出DNA,根据萃取出的DNA可以知道土壤微生物的集群结构。所以使用本试剂盒萃取出的土壤DNA能够使用PCR-DGGE解析出土壤微生物的集群结构以及土壤诊断、土壤DNA的定量,从而知道土壤中生物的数量

       ISOIL Large For Beads VER2是能够从5g土壤样本中萃取出DNA的试剂盒。将界面活性剂进行的化学性溶菌和Beads进行的物理性菌体破碎 共同使用,能够高效地萃取出土壤中的DNA。除了萃取的基本步骤,还有再提纯的另外两种步骤可选,使用范围更广。

◆概述


赖氨酰内肽酶最早从土壤细菌无色杆菌Achromobacter lyticus中分离得到,能够特异性识别与切割肽链中Lys羧基端肽键,常被用于蛋白测序及Lys-X化合物的酶催化合成。

艾美迪选用Achromobacter lyticus来源的,克服大肠杆菌重组表达技术难题进行制备。本品最适pH 9.0~9.5,等电点为pH 6.9~7.0;最适反应温度30~37℃,50℃以上稳定性下降。在适宜温度下,该酶稳定性良好,在4 mol/L尿素或0.2%SDS溶液中30℃孵育6 hr后,生物活性未发生降低。可广泛用于蛋白组学分析,蛋白、抗体类药物质量检测,多肽、蛋白类产品制备工艺中。

重组 E. coli 菌株,其克隆有来自的无色杆菌(A. lyticus蛋白酶I基因。

 


◆酶活定义


0.2 mol/L pH 9.0Tris-HCl缓冲中,温度为37±2℃的条件下, 1 min催化底物AC-Lys-PNA生成1 μmol PNA所需的酶量为一个活性单位(AU)。

将 rLys-C以DTT 还原处理后, 还原电泳显示纯度>99%

本品为冻干粉,使用前先使用超纯水或25 mmol/L Tris-HCl,pH 8.0~8.5溶解。

  免疫亲和层析柱【Smart Column系列】用于提取食品中的黄曲霉毒素和赭曲霉毒素。以前在免疫亲和柱上样前处理为必须用有机溶剂将实验样本提取液稀释到2%水平,但是那样提取液会变浑浊而很难分析调味品。

  本产品则是在原产品的基础上提高了有机溶剂高耐性。

特点

1.因具有有机溶剂高耐性,少量的样本溶液即可快速提取。

2.黄曲霉毒素提取通常使用乙腈和甲醇,【AFLAKING】对于两者具备高耐性。乙腈浓度调制到20%,甲醇浓度调制到40%,可保证黄曲霉毒素的回收率在80%以上。大幅缩短了前处理时间,提高分析效率。

3.可正确分析以前分析困难的调味品类。

  黄曲霉毒素具有毒性,在多种食品和饲料中被检测出来。截止到现在,发现了多种黄曲霉毒素的相关化合物。对于食品卫生问题,黄曲霉毒素B1/B2/G1/G2/M1中尤其是黄曲霉毒素B1是天然物中致癌性最高的。


◆赭曲霉毒素提取用免疫亲和柱(OCHRAKING-SmartColumn系列)


特点:

1.具有有机溶剂高耐性,可提取少量提取液中的赭曲霉毒素。

2.大幅缩短前处理时间,提高分析效率。

(ADP-GK)
(2) ADP + D-glucose → G-6-P + AMP

(glucose-6-phosphate dehydrogenase)
(3) G-6-P + NAD+ → gluconate-6-phosphate + NADH + H+

 Q1. Should the pH of the sample be adjusted even for samples in acidic media?

The pH of the assay solution after the sample is added should be the same as that of the assay buffer that is supplied with the kit.
Low sample volumes (e.g. 0.1 mL) are not likely to affect the pH of the assay solution and therefore may not require pH adjustment.
Samples above 0.1 mL are more likely to affect the pH of the assay solution and therefore the pH of these samples should be adjusted as described in the data booklet, prior to addition to the assay.

Q2. Is the K-GCROLGK Assay Kit suitable for measurement using cell culture media samples?

Yes, assuming that the concentration of the analyte in the sample (after sample preparation) is above the limit of detection for the kit.  It may be sufficient to use the sample directly in the assay after clarification by centrifugation / filtering followed by dilution (if required) in distilled water. 

Q3. Is K-GCROLGK specific for glycerol?

K-GCROLGK is highly specific for glycerol.
Some compounds that are known not to react or interfere with the assay include:
Polyethylene glycol
Ethylene glycol
Propylene glycol 

Q4. Sometimes a negative absorbance change is obtained for the blank samples, is this normal? Should the real value (negative absorbance change) or “0” be used in the calculation of results?

Sometimes the addition of the last assay component can cause a small negative absorbance change in the blank samples due to a dilution effect and in such cases it is recommended that the real absorbance values be used in the calculation of results.

Q5. There is an issue with the performance of the kit; the results are not as expected.

If you suspect that the Megazyme test kit is not performing as expected such that expected results are not obtained please do the following:

  1. Ensure that you have tested the standard sample that is supplied with the Megazyme test kit.
  2. Send the results of the kit standard, blank samples and the results obtained for your sample, in the relevant MegaCalc spreadsheet (if available) to Megazyme (cs@megazyme.com). Where available the relevant MegaCalc spreadsheet can be downloaded from where the product appears on the Megazyme website.
  3. State the kit lot number being used (this is found on the outside of the kit box).
  4. State which assay format was used (refer to the relevant page in the kit booklet if necessary).
  5. State exact details of any modifications to the standard procedure that is provided by Megazyme.
  6. State the sample type and describe the sample preparation steps if applicable.

Q6. The pH of my sample is low (pH ~ 3.0), do I need to adjust this before I use the sample in the kit assay?

The final pH of the kit assay after the sample is added should not change from what it should be (as stated in the kit for the assay buffer). If it does change then the sample will require pH adjustment. In most cases the sample volume being used is low relative to the final assay volume and in this case the pH of the kit assay is unlikely to be affected.

Q7. How can I work out how much sample to extract and what dilution of my sample should be used in the kit assay?

Where the amount of analyte in a liquid sample is unknown, it is recommended that a range of sample dilutions are prepared with the aim of obtaining an absorbance change in the assay that is within the linear range.
Where solid samples are analysed, the weight of sample per volume of water used for sample extraction/preparation can be altered to suit, as can the dilution of the extracted sample prior to the addition of the assay, as per liquid samples.

Q8. Can you explain, step by step, how to follow the method and perform the kit assay?

For users who are not familiar with how to use the Megazyme tests kits then it is recommended that they follow this example, e.g. D-Fructose/D-Glucose Assay kit K-FRUGL (http://secure.megazyme.com/D-Fructose-D-Glucose-Assay-Kit):

1. The kit components are listed on pages 2-3 of the kit booklet.
2. Prepare the kit reagents as described on page 3.
3. For separate measurements of glucose and fructose follow procedure A on page 4.
4. Pipette the volumes listed for water, sample, solution 1 and solution 2 into 3 mL, 1 cm pathlength cuvettes. Duplicate sample assays and duplicate blanks are recommended. Mix the contents of each cuvette by inversion (seal the cuvette using parafilm or a plastic cuvette cap – do not use a finger) then after ~3 min record the first absorbance reading of each cuvette at 340 nm (this is reading A1).
5. Then add suspension 3 and mix the contents of each cuvette by inversion. Incubate for 5 minutes then record the absorbance reading of each cuvette at 340 nm (this is reading A2). NB. It is essential that the reaction is compete. To assess this, record the absorbances at ~ 2 minute intervals and until the absorbance plateaus. A stable absorbance indicates that the reaction is complete. If the absorbance continues to increase then continue to record absorbances until it plateaus and only then record absorbance reading A2.
6. Then add suspension 4 and mix the contents of each cuvette by inversion. Incubate for 5 minutes then take absorbance reading of each cuvette at 340 nm (this is reading A3). NB. As above, assess that the reaction has completed by take subsequent readings at ~2 min intervals.
7. For simple, automated results analysis, input the absorbance readings (A1, A2, A3) for samples and blanks into the 
K-FRUGL MegaCalc.

To ensure that the assay is working, and being performed correctly it is recommend that the test is performed using the standard sample that is provided with the kit and to obtain the expected values before proceeding to test real samples.
It is recommend that new users also watch 
this video which highlights how to perform the assays.
Many of the other Megazyme test kits follow a similar format.

Q9. I have some doubts about the appearance/quality of a kit component what should be done?

If there are any concerns with any kit components, the first thing to do is to test the standard sample (control sample) that is supplied with the kit and ensure that the expected value (within the accepted variation) is obtained before testing any precious samples. This must be done using the procedure provided in the kit booklet without any modifications to the procedure. If there are still doubts about the results using the standard sample in the kit then send example results in the MegaCalc spread sheet to your product supplier (Megazyme or your local Megazyme distributor).

Q10. Can the sensitivity of the kit assay be increased?

For samples with low concentrations of analyte the sample volume used in the kit assay can be increased to increase sensitivity. When doing this the water volume is adjusted to retain the same final assay volume. This is critical for the manual assay format because the assay volume and sample volume are used in the calculation of results.

Q11. How much sample should be used for the clarification/extraction of my sample?

The volume/weight of sample and total volume of the extract can be modified to suit the sample. This will ultimately be dictated by the amount of analyte of interest in the sample and may require empirical determination. For low levels of analyte the sample:extract volume ratio can be increased (i.e. increase the sample and/or decrease the total extraction volume).

Alternatively, for samples with low concentrations of analyte, a larger sample volume can be added to the kit assay. When altering the sample volume adjust the distilled water volume added to the assay accordingly so that the total assay volume is not altered.

Q12. Can the test kit be used to measure biological fluids and what sample preparation method should be used?

The kit assay may work for biological fluids assuming that inositol is present above the limit of detection for the kit after any sample preparation (if required). Centrifugation of the samples and use of the supernatant directly in the kit assay (with appropriate dilution in distilled water) may be sufficient. The test kit has not been tested using biological fluids as samples because it is not marketed or registered as a medical device. This will therefore require your own validation.

Q13. Can the manual assay format be scaled down to a 96-well microplate format?

The majority of the Megazyme test kits are developed to work in cuvettes using the manual assay format, however the assay can be converted for use in a 96-well microplate format. To do this the assay volumes for the manual cuvette format are reduced by 10-fold. The calculation of results for the manual assay format uses a 1 cm path-length, however the path-length in the microplate is not 1 cm and therefore the MegaCalc spreadsheet or the calculation provided in the kit booklet for the manual format cannot be used for the micropalate format unless the microplate reader being used can.

There a 3 main methods for calculation of results using the microplate format:

  1. The easiest method is to use a microplate reader that has a path-length conversion capability (i.e. the microplater reader can detect the path-length of each well and convert the individual readings to a 1 cm path-length). This will allow values to be calculated using the MegaCalc calculation software which can be found where the product is located on the Megazyme website.
  2. Perform a standard curve of the analyte on each microplate that contains test samples and calculate the result of the test samples from the calibration curve (concentration of analyte versus absorbance).
  3. Perform a standard curve of the analyte in both the cuvette format (i.e. with a 1 cm path-length) and the 96-well microplate format and use these results to obtain a mean conversion factor between the cuvette values and the microplate values. Subsequent assays in the microplate format can then be converted from the calculated conversion factor.

Q14. When using this kit for quantitative analysis what level of accuracy and repeatability can be expected?

The test kit is extremely accurate – at Megazyme the quality control criteria for accuracy and repeatability is to be within 2% of the expected value using pure analytes.

However, the level of accuracy is obviously analyst and sample dependent.

Q15. Is it possible to add a larger volume then 2 μL of enzyme to the microplate assay? In some instances 2 μL can be difficult to pipette manually.

Yes, instead of adding 2 μL of enzyme suspension an alternative is to dilute the enzyme and add a larger volume to the microplate assay.

Dilute the assay buffer 10-fold with distilled water and use this as the diluent to dilute an aliquot of the enzyme suspension also by 10-fold. Instead of 2 μL, use 20 μL of the diluted enzyme in the microplate assay.

Q16. Must the minimum absorbance change for a sample always be at least 0.1?

No. The 0.1 change of absorbance is only a recommendation. The lowest acceptable change in absorbance can is dictated by the analyst and equipment (i.e. pipettes and spectrophotometer) and therefore can be can be determined by the user. With accurate pipetting, absorbance changes as low as 0.02 can be used accurately.
If a change in absorbance above 0.1 is required but cannot be achieved due to low concentrations of analyte in a sample, this can be overcome by using a larger sample volume in the assay to increase the absorbance change and thereby increase sensitivity of the assay. When doing this the increased volume of the sample should be subtracted from the distilled water volume that is added to the assay so that the total assay volume is unaltered. The increase sample volume should also be accounted for when calculating final results. 

Q17. Can the sensitivity of the kit assay be increased?

Yes. Samples with the lower concentrations of analyte will generate a lower absorbance change. For samples with low concentrations of analyte, a larger sample volume can be used in the assay to increase the absorbance change and thereby increase sensitivity of the assay. When doing this the increased volume of the sample should be subtracted from the distilled water volume that is added to the assay so that the total assay volume is unaltered. The increase sample volume should also be accounted for when calculating final results.

Q18. To measure fermentation samples contain high microbial cell density, is cell disruption required?

Cell disruption is only require to measure glycerol within microbial cells.

To measure glycerol in the extracellular media only then cell disruption is not required and centrifugation of the sample may be sufficient, e.g.:

(a) Determination of glycerol in cell culture media/supernatants. In general, the concentration of glycerol in cell culture media/supernatants can be determined without any sample treatment (except clarification by centrifugation/filtering or dilution according to the dilution table, if necessary). Typically, no clarification or dilution is required, and a sample volume of 0.1 mL is satisfactory.

If interference is suspected then sample clarification/deproteinisation using carrez reagents or perchloric acid should be used (methods are provided in the kit booklet).

APSC主要提供有机凝胶色谱标准品(Organic GPC/SEC Standards)和水性凝胶色谱标准品(Aqueous GPC/SEC Standards)两类产品。
GPC(Gel Permeation Chromatography),凝胶渗透色谱,又称为尺寸排阻色谱(Size Exclusion Chromatography,简称SEC),它是基于体积排阻的分离机理,通过具有分子筛性质的固定相,用来分离相对分子质量较小的物质,并且还可以分析分子体积不同、具有相同化学性质的高分子化合物。
GPC是一种特殊的液相色谱,所用仪器与高效液相色谱仪类似,GPC的分离是利用体积排除机理,装填的是多孔性凝胶或微粒,孔径大小与待分离的聚合物分子相似,体积大的高分子化合物不能进入凝胶孔,最先从凝胶粒间流出,淋出体积(时间)最小,高聚物依分子量从大到小依次淋出。这样,通过做已知分子量的高分子聚合物的标准曲线,就能分析同系未知待测高分子化合物了。

● 接触溶液的表面进行了氟化处理(氟化层),具有优越的抗挥发性(尤其对有机溶剂)和耐药性。

● 采用氟化处理技术,经过氟化处理的广口瓶表面效果持久。

 (若表层破损,基本材料PE外露,氟化处理就会失效,使用时请注意。)

● 广口瓶的基本材料为PE,价格优惠,瓶身轻巧。

● 经过氟化处理的产品都标有批号,方便进行生产管理。

使用前请使用刺激性较小的温水或清洗液进行清洗。

请勿使用清洗刷或含研磨剂的去污粉等,以免损伤氟化层。
